PYY is released from the small intestine after eating and inhibits hunger signals from the hypothalamus.

Insulin also plays an important role in weight regulation by signaling cells to store or release sugar from the blood into our cells for energy production. The amount of sugar in the blood affects our hunger signals.

When there is too much sugar in the blood, insulin tells cells to store it so it cannot be used for energy production. When there is not enough sugar in the blood, insulin tells cells to release glucose into the bloodstream for energy production. This cycle ‘primes’ the body for hunger or satiety, which can be affected by meal timing and composition.

Leptin’s effects are distinct from the other two hormones’ effects; leptin signaling acts more directly on the hypothalamus to suppress appetite. Ghrelin stimulates appetite by acting on ghrelin receptors in the hypothalamus. PYY also inhibits appetite like leptin but works through a different mechanism to suppress appetite.

Hypothalamic neuropeptide Y (NPY) is involved in regulating feeding behavior and metabolism. NPY induces hunger by stimulating hypothalamic neurons that ultimately target the brainstem to increase food intake. NPY expression increases with fasting or caloric restriction, contributing to increased energy demand during times of low caloric intake.

The conclusion that ghrelin is most important for weight loss is a bit misleading since all three hormones play a role in weight loss. Leptin and PYY are known to suppress appetite, while ghrelin stimulates appetite. Ghrelin has been shown to have more of an effect on weight regulation than leptin or PYY, but more research is needed to determine if it is the most important hormone for weight loss.
